Marmagkiolis completed his medical education at Aristotelian University of Thessaloniki in Greece. He later earned an MBA from the Open University of Cyprus and completed the Global Clinical Scholars Research Training Program at Harvard Medical School in Boston. Born in Kavala, Greece, he is married with four children, is fluent in Greek, English, and French, and holds both United States and Greek citizenship.
